Second dose of Johnson & Johnson vaccine dramatically increased protection against Covid-19, company announcement Tuesday morning.

In a clinical trial, researchers found that two doses of the vaccine offered 94% efficacy against mild to severe Covid-19 in the United States, against 74 percent conferred in one fell swoop, the company reported. And two injections have shown 100 percent effectiveness against serious illness, although that estimate has a wide range of uncertainties.

The data, presented in a press release, has been submitted to the Food and Drug Administration, Johnson & Johnson said. Since the company received an emergency clearance in February, 14.6 million people in the United States received its single injection vaccine.

Friday, an FDA advisory committee advised that the agency authorize Pfizer-BioNTech recalls for vaccine recipients who are at least 65 years of age or at high risk of Covid. This vaccine, like Moderna’s, offers high levels of initial protection after two doses, which then appear to decrease slightly over several months.

In contrast, Johnson & Johnson’s vaccine showed little sign of decline. Researchers published a study last week, comparing 390,517 people vaccinated to 1,524,153 unvaccinated. Up to five months after vaccination, the efficacy of the Johnson & Johnson vaccine against hospitalization remained stable at around 81%.

As the pandemic unfolded, people who received the Johnson & Johnson vaccine have waited for advice to find out if they will need a reminder. The new clinical trial, which recruited 32,000 volunteers worldwide, compared people who received one dose of Johnson & Johnson to those who received two doses eight weeks apart.

The researchers found that the second stroke raised the level of antibodies in the volunteers’ blood to four times the level produced by the first stroke. This improvement resulted in enhanced protection.

A lot of people had their Johnson & Johnson drawn well over eight weeks ago. Other research suggests that the extra time between doses could mean even better protection.

In a separate study announcement Johnson & Johnson last month gave clinical trial volunteers boosters six months after their first dose and then measured their antibody levels.

Initially, the researchers reported that the antibodies rose nine times higher than after the first dose. But in Tuesday’s press release, the company said the level has continued to rise, reaching 12 times higher than initial levels.

Some preliminary studies to suggest that higher levels of antibodies against the coronavirus produce higher levels of protection against Covid. If this is true, then a second injection of Johnson & Johnson given after waiting for several months may prove to be even more effective than after just eight weeks.
